Phytoestrogenic Potential of Resveratrol by Selective Activation of Estrogen Receptor-α in Osteoblast Cells

چکیده

Abstract Resveratrol obtained in grape seed and skin is structurally similar to a synthetic estrogen diethylstilbestrol. The endogenous estrogen, 17β-estradiol, induces cellular responses by binding the receptor alpha beta. bone fracture due decreased mineral density postmenopausal women linked reduced estrogen. adverse drug reactions of hormone replacement therapy warrant identifying unique natural compounds with ER-subtype specificity improve health. considered phytoestrogen; however, its isoform selectivity has not yet been established on osteoblast cell lines. Therefore, vitro silico docking studies were performed analyze affinity resveratrol towards β-isoforms. was evaluated for actions proliferation differentiation primary rat calvarial osteoblasts marrow cells. Osteoblasts specifically increased expression cells; there no effect beta expression. In further confirmed specificity. observed differences orientation, interaction pattern, at active site are supported western blot analysis. mimetic action suggests therapeutic potential as anabolic agent osteoporosis.

Receptor in Breast Cancer Cells Expressing Wild-Type Estrogen -Selective Estrogen Receptor Modulator Complex α Receptor Gene Expression Profiles with Activation of the Estrogen

Selective Estrogen Receptor Modulators (SERMs) are a new class of drugs that bind to estrogen receptor (ER) and elicit agonistic or antagonistic responses, depending on the target tissue. We have developed an in vitro system in which some SERMs (4-hydroxytamoxifen and resveratrol) demonstrate estrogenic response through wild-type (wt) ER, whereas others (raloxifene and GW7604) remain antiestrog...

Antagonists Selective for Estrogen Receptor α.

To develop compounds that are antagonists on ER , but not ER , we have added basic side-chains typically found in nonsteroidal antiestrogens to pyrazole compounds that bind with much higher affinity to ER than to ER . In this way we have developed basic side-chain pyrazoles (BSC-pyrazoles) that are high affinity, potent, selective antagonists on ER . These BSCpyrazoles are themselves inactive o...

Estrogen Receptor α Regulates Dlx3-Mediated Osteoblast Differentiation.

Estrogen receptor α (ER-α), which is involved in bone metabolism and breast cancer, has been shown to have transcriptional targets. Dlx3 is essential for the skeletal development and plays an important role in osteoblast differentiation. Various osteogenic stimulators and transcription factors can induce the protein expression of Dlx3. However, the regulatory function of ER-α in the Dlx3 mediat...

Breast Cancer Cells by Histone Deacetylase Inhibition in Human α Transcriptional Activation of Estrogen Receptor

Recent findings have established a connection between DNA methylation and transcriptionally inactive chromatin characterized by deacetylated histones. Because the absence of estrogen receptor a (ERa) gene expression has been associated with aberrant methylation of its CpG island in a significant fraction of breast cancers, we tested whether histone deacetylase activity contributes to the transc...
